Cientistas of Instituto of Tecnologia of Shibaura, in Japão, created enhanced vitamin K compounds capable of regenerating lost neurons and combating neurodegenerative diseases such as Alzheimer and Parkinson. The new analogues demonstrated three times greater activity in transforming neural progenitor cells into functional neurons compared to natural vitamin K, according to a study published in July 2025 in the journal ACS Chemical Neuroscience.
The research represents a significant advance in brain regenerative medicine. Enquanto Current medications only alleviate symptoms, the new approach seeks to restore damaged brain tissue and replace lost neurons — an objective previously considered distant in the treatment of neurodegenerative diseases.
Vitamina K beyond blood clotting
Vitamin K is traditionally known for its effects on blood clotting and bone health. Nos In recent years, however, researchers have also linked it to brain protection and neuronal differentiation — the biological process by which immature neural cells transform into fully functional neurons.
The natural form of vitamin K found in the body is menaquinone 4 (MK-4). Contudo, its isolated effects were not sufficiently potent for future applications in regenerative medicine aimed at neurodegenerative diseases. Essa limitation motivated the work of Instituto, Tecnologia, and Shibaura researchers.
Professor Associado Yoshihisa Hirota and Professor Yoshitomo Suhara led the project that synthesized 12 hybrid analogues of vitamin K. The strategy consisted of combining vitamin K with components related to vitamin A, specifically retinoic acid — a substance already known to promote neuronal differentiation.
Compostos hybrids with tripled activity
The researchers created hybrid molecules that preserved the biological activity of both vitamin K and retinoic acid. One compound in particular stood out among the 12 synthesized: Análogo of Vitamina K Inovador (Novel VK), which combined the retinoic acid backbone with a methyl ester side chain.
Tests on mouse neural progenitor cells revealed impressive results:
- Novel VK showed neuronal differentiation activity 3 times higher than the natural control
- Superou significantly reduces the effects of conventional vitamin K compounds
- Aumentou robust neuronal growth markers (Map2 protein)
- Converteu turns into bioactive MK-4 with greater efficiency within cells
The researchers also measured microtubule-associated protein 2 (Map2), a marker directly related to neuronal growth and development. Novel VK demonstrated significantly stronger activity in this parameter compared to natural compounds.
Mecanismo action via glutamate receptors
The team investigated the biochemical pathways responsible for the neuroprotective effects. Gene expression analysis in neural stem cells pointed to metabotropic glutamate receptors (mGluRs), structures that appear to fundamentally contribute to vitamin K-induced neuronal differentiation through epigenetic and transcriptional regulation.
The effect was specifically associated with mGluR1. Essa discovery has particular relevance because mGluR1 was already linked in scientific literature to synaptic transmission — communication between neurons. Genetically modified Camundongos lacking mGluR1 exhibit motor and synaptic problems that overlap with the types of dysfunction seen in neurodegenerative diseases such as Alzheimer and Parkinson.
Estudos molecular docking techniques revealed that the new vitamin K compound showed higher binding affinity to mGluR1 than natural MK-4, strengthening the hypothesis of its improved mechanism of action.
Travessia of the blood-brain barrier
A critical challenge in medicines for brain diseases is the ability to cross the blood-brain barrier — a biological structure that protects the brain but prevents the passage of many substances. Experimentos with mice demonstrated that Novel VK:
- Atravessou efficiently crosses the blood-brain barrier
- Produziu MK-4 concentrations in the brain higher than those in the control group
- Apresentou stable pharmacokinetic profile during testing
- Converteu into MK-4 easier than natural vitamin K
Esses results indicate that the compound is not only more potent in vitro, but can also reach the brain target in biologically significant quantities — an essential requirement for any future neurological therapeutics.
Perspectivas to Alzheimer, Parkinson and Huntington
Neurodegenerative diseases such as Alzheimer, Parkinson and Huntington progressively damage the brain, destroying neurons — the cells responsible for transmitting messages in the nervous system. As these cells are lost, patients develop memory problems, cognitive decline, and movement difficulties that often become severe enough to require constant care.
Currently approved drugs, including the anti-amyloid therapies lecanemab and donanemab (recognized by the FDA), can slow progression in patients in the early stages, but they do not restore lost memories or rebuild damaged brain tissue. Nova VK represents a paradigm shift: instead of simply slowing degeneration, it seeks to induce active regeneration of neurons.
The Dr. Hirota said the research offers a potentially innovative approach. A vitamin K-derived drug that slows the progression of Alzheimer or improves its symptoms would not only elevate the quality of life of patients and families, but also significantly reduce the growing burden of health and long-term care spending on public and private systems.
Caminho long to the clinic
Importante emphasize that the findings, although promising, are based on cellular studies and experiments with mice, not clinical trials with humans. Nenhum vitamin K-derived medication has thus far been shown to repair the brains of people with Alzheimer, Parkinson, or Huntington disease.
The expectation is that this line of research will gradually advance from laboratory results to clinically significant treatments. The mGluR1 pathway identified in this study provides researchers with a clear molecular target for developing future brain repair therapies — an essential step in the long process from fundamental discovery to approved medicine for patients.
